Modifikasi Screen Printed Carbon Electrode Menggunakan Cerium dan Optimasi Kondisi Percobaan Biosensor Elektrokimia untuk Deteksi DNA Mitokondria Sus Scrofa

The use of cerium in sensor applications has been widely develop. In this research, electrochemical
biosensors have been developed to support Sus scrofa mitochondrial DNA using SPCE-Ce. The surface of
the SPCE electrode before and after an electrodeposition with cerium, was characterized using SEM and
cyclic voltammetry (CV). The design of pork nucleotide sequence as DNA probes, was determined using
NCBI blast program and T-Coffee. The DNA probe was immobilized to SPCE-Ce trough electrostatic
interactions between the surface of cerium and the DNA phosphate backbone. The probe-target DNA
hybridization was characterized using DPV. The result of the characterization using SEM showed a flat and
uniform of electrode surface for SPCE-Ce than the bare SPCE surface. The voltammogram cyclic shows an
increase in the peak of the ferrycianide current for SPCE-Ce. The result of the DNA probe sequence design
are 5’-TATTIATACCAATCACTAIC-3’ which shows 100% homology with pig DNA and 0% homology with
chicken and beef DNA. The linear equation of target concentration is I = 0.0303 [target DNA] + 3.947 for
the target DNA concentration range 5 – 30 µg/mL, and the LOD value is 1.44 µg/mL. This biosensor can be used to determine pig mtDNA concentration in the real samples
